Carica papaya L., Caricaceae is cultivated almost all over tropical countries. Quercetin is a flavonoid in papaya leaves with
many biological activities. This study was designed for isolation of quercetin from papaya leaves. Fresh papaya leaves were
collected in Dong Nai province. The isolation and purification of flavonoid compounds from Papaya leaves was performed
using the extraction methods with ethanol, shaking liquid-liquid distribution and vacuum column chromatography. The structure of pure compounds was determined using the spectrophotometric methods. Results showed that 11 subfractions (F1-F11) were isolated from ethyl acetate fractions using vacuum column chromatography. After purification of F9 fraction, 50 mg of CP2 was collected with 99.01% purity by HPLC-DAD method. The structure of CP2 compound was determined to be quercetin using spectroscopic methods including uV, IR, ESI-MS and 1D-NMR, 2D-NMR. In conclusion, quercetin isolated from Carica papaya leaves can be used as a reference substance for assessing ingredients or medicinal preparations containing papaya leaves.
